目的了解丰台区流行的麻疹野病毒的基因特征,为控制和消除麻疹提供依据。方法采集2013年春季疑似麻疹爆发和散发患者咽拭子标本分离病毒,通过逆转录一聚合酶链反应(RT-PCR),从分离到的麻疹病毒中扩增出核蛋白(nucleoprotein,N)基因C末端634个核苷酸片段,再对扩增产物进行核苷酸序列测定和分析,并以C末端450个核苷酸片段构建基因亲缘关系树,进行遗传距离分析。结果共分离麻疹病毒39株,测序后为Hl基因型(28株)和D8基因型(11株)。结论丰台区2013年春季麻疹流行株仍属于H基因组,Hl基因型,Hla亚型。此外北京市丰台区首次发现D8基因型麻疹野病毒。
Objective To understand the genetic characteristics of the wild measles wild virus in Fengtai District and provide the basis for controlling and eliminating measles. Methods Viruses were isolated from the throat swabs in the spring of 2013 and the throat swab specimens were collected. The nucleoprotein (N) gene was amplified from the isolated measles virus by reverse transcription-polymerase chain reaction (RT-PCR) C-terminal 634 nucleotide fragments, and then the amplified products were sequenced and analyzed. The C-terminal 450 nucleotide fragments were used to construct the genetic relationship tree for genetic distance analysis. Results A total of 39 strains of measles virus were isolated. After sequencing, Hl genotype (28 strains) and D8 genotype (11 strains) were sequenced. Conclusion The epidemic strains of measles in Fengtai district in spring 2013 still belonged to H genotype, Hl genotype and Hla subtypes. In addition, Fengtai District of Beijing found for the first time D8 genotype of measles wild virus.
