Vav1是造血系统中的重要蛋白,具有鸟苷酸交换因子(guanine nucleotide exchange factor,GEF)活性,能够催化Rac/Rho GTP酶的活性,从而参与众多的细胞事件,如细胞骨架重组,基因的转录,免疫细胞的发育和活化,细胞增殖和凋亡等.近年来,在包括乳腺癌在内的人类癌症中均发现有Vav1的表达,但对于Vav1在乳腺癌细胞中的具体功能和机制仍不清楚.本研究中,通过慢病毒包装系统感染乳腺癌细胞系T47D构建了稳定表达Vav1的乳腺癌细胞系,通过WST-1检测细胞增殖和细胞流式法检测细胞周期发现Vav1能够促进细胞增殖,同时通过蛋白免疫印迹技术和报告基因系统发现这一过程可能跟Vav1增强转录因子NF-κB转录活性上调细胞周期相关蛋白Cyclin D1相关.

Vav1 was found to promote cell proliferation, At the same time, we found that this process may be related to the up-regulation of cyclin D1, a transcription factor of NF-κB by Vav1, by Western blot and reporter system.
