目的:探讨多潘立酮(DMP)-卡波姆胃内黏附微丸的制备方法,同时评价其体外释放行为与黏附性能。方法:卡波姆、聚维酮(PVP)、羟丙基纤维素(HPC)为黏附材料,30%的氯化钠水溶液作为润湿剂制备软材,采用挤出-滚圆法制备多潘立酮-卡波姆胃内黏附微丸。以体外释放度与黏附性作为评价指标,利用正交设计试验优选最佳处方并进行验证与评价。结果:正交设计方案结果显示卡波姆、羟丙基纤维素、PVP质量比为80:80:15时所得的处方最佳,此时的多潘立酮-卡波姆胃内黏附微丸所得黏附与缓释效果最佳。结论:利用挤出-滚圆法制备多潘立酮-卡波姆胃内黏附微丸,有着工艺简便,同时重现性良好等优势,对于工业化的生产十分适用。
Objective: To investigate the preparation of domperidone (DMP) - carbomer gastric adhesion pellets, and to evaluate the in vitro release behavior and adhesion properties. Methods: Carbomer, polyvinylpyrrolidone (PVP) and hydroxypropylcellulose (HPC) were used as adhesive materials, 30% sodium chloride aqueous solution was used as wetting agent to prepare soft materials. The extrusion-spheronization method was used to prepare domperidone- Pom stomach adhesion pellets. In vitro release and adhesion as evaluation index, orthogonal design test to optimize the best prescription and validation and evaluation. Results: The results of orthogonal design showed that the optimal formulation was obtained when the weight ratio of Carbomer, hydroxypropylcellulose and PVP was 80:80:15. At this time, the adhesion between the domperidone-carbomer gastric pellet and Slow release the best. Conclusion: The extrusion-spheronization method for the preparation of domperidone-carbopol gastric adhesion pellets has the advantages of simple process and good reproducibility, and is very suitable for industrialized production.
