目的 :探讨川芎嗪对SD大鼠被动型Hermann肾炎 (PHN)的蛋白尿、血肌酐、血栓素A2 (TxA2 )、前列环素I2 (PGI2 )的代谢产物——— 6 Keto PGF1α及肾脏病变的影响。方法 :给大鼠静脉注射兔抗鼠肾小管上皮抗原 (Tub Ag)的抗血清建立PHN模型 ,此后 ,每两天给大鼠腹腔注射川芎嗪直到第五周止。在此期间 ,定期用磺基水杨酸、苦味酸和放射免疫法分别测定大鼠蛋白尿、血肌酐、TxA2 和 6 Keto PGF1α的含量 ,另用光镜、电镜和直接免疫荧光染色 (兔IgG和鼠IgG)对其肾脏病理学变化进行检查。结果 :用川芎嗪处理的PHN大鼠 ,其尿蛋白、血肌酐、尿液中TxA2 和肾组织病变均较未处理的PHN大鼠明显减少或减轻 ,而尿液中的 6 Keto PGF1α则较未处理的PHN大鼠有所提高。结论 :川芎嗪能减轻PHN大鼠的蛋白尿、血肌酐、TxA2 和肾脏病变。提高 6 Keto PGF1α的水平。上述结果提示 :川芎嗪能改善PHN大鼠TxA2 和PGI2 平衡 ,可用于膜性肾小球肾炎的防治。
Objective: To investigate the effects of ligustrazine on proteinuria, serum creatinine, TxA2, and PGI2 metabolites in idiopathic hermann nephritis (PHN) --- 6 Keto PGF1α and renal lesions influences. METHODS: PHN models were established by intravenous injection of rabbit anti-mouse anti-mouse anti-mouse renal tubular epithelial antigen (Tub Ag). Thereafter, tetramethylpyrazine was intraperitoneally injected to the rats every 5 days until the 5th week. During this period, the contents of proteinuria, serum creatinine, TxA2 and 6 Keto PGF1α in rats were measured regularly with sulfosalicylic acid, picric acid and radioimmunoassay, and the contents of serum creatinine, TxA2 and Keto PGF1α were detected by light microscopy, electron microscopy and direct immunofluorescence staining And murine IgG) examined their renal pathological changes. Results: Compared with untreated PHN rats, the urinary protein, serum creatinine, urine TxA2 and renal tissue lesions in PHN rats treated with Ligustrazine were significantly reduced or decreased, while the urinary 6 Keto PGF1α was less PHN rats treated increased. Conclusion: Tetramethylpyrazine can reduce proteinuria, serum creatinine, TxA2 and renal lesions in PHN rats. Increase the level of 6 Keto PGF1α. The above results suggest that tetramethylpyrazine can improve the balance of TxA2 and PGI2 in PHN rats and can be used in the prevention and treatment of membranous glomerulonephritis.
