目的:探讨中药复方固本防哮饮对哮喘缓解期小鼠白细胞介素-17A(IL-17A)、白细胞介素-10(IL-10)及CXC趋化因子配体10(CXCL10)表达的影响。方法:采用卵蛋白(OVA)致敏和呼吸道合胞病毒(RSV)滴鼻诱导激发BALB/c雌性小鼠,建立哮喘缓解期动物模型,随机分为正常组,模型组,固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组。用ELISA法和Real-time PCR法分别检测肺组织中IL-17A、IL-10及CXCL10的表达水平。结果:模型组小鼠肺组织中IL-17A、CXCL10表达均显著高于正常组(P<0.01),而IL-10表达显著低于正常组(P<0.01);固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组小鼠肺组织中IL-17A、CXCL10表达均低于模型组(P<0.05),而IL-10表达高于模型组(P<0.05)。结论:固本防哮饮治疗哮喘的作用机制可能是通过上调IL-10水平,降低IL-17A、CXCL10水平,减轻气道炎性反应,从而达到防治哮喘的目的。
Objective: To investigate the effect of Guben anti-asthma drink on the expression of interleukin-17A, interleukin-10 and CXCL10 in mice with asthma remission influences. Methods: BALB / c female mice were induced by ovalbumin (OVA) sensitization and respiratory syncytial virus (RSV) nasal instillation. Animal models of asthma remission were established and randomly divided into normal group, model group, , Middle and high dose group, montelukast sodium group and dexamethasone group. The expression of IL-17A, IL-10 and CXCL10 in lung tissue were detected by ELISA and Real-time PCR respectively. Results: The expression of IL-17A and CXCL10 in model group was significantly higher than that in normal group (P <0.01), while the expression of IL-10 in model group was significantly lower than that in normal group (P <0.01) The expression of IL-17A and CXCL10 in the middle and high dose groups, the montelukast sodium group and the dexamethasone group were lower than those in the model group (P <0.05), while the expression of IL-10 in the model group was higher than that in the model group <0.05). Conclusion: The mechanism of Guben anti-asthma drink in treating asthma may be to prevent and treat asthma by up-regulating the level of IL-10, decreasing the levels of IL-17A and CXCL10 and alleviating the airway inflammatory reaction.
