GM-CSF在Ⅱ型登革病毒E蛋白免疫中的作用

  目的:探讨粒细胞-巨噬细胞集落刺激因子(GM-CSF)作为免疫佐剂对登革病毒E蛋白免疫保护的影响,评估GM-CSF作为免疫佐剂的多面性。方法 实验共设3组,分别为E蛋白组、E蛋白+pCAG-GM组(表达GM-CSF)和对照组,采用肌肉注射和足垫注射两种方式进行免疫。分别利用ELISA和PRNT检测血清总IgG水平和中和抗体水平,ELISPOT检测细胞因子的分泌,攻毒实验检测动物保护率。结果 ELISA和PRNT结果显示E蛋白组和E蛋白+pCAG-GM组小鼠血清中总IgG水平、中和抗体水平均在免疫后有一定的升高,但两组之间无显著差异;ELISPOT结果显示E蛋白+pCAG-GM组的IFN-γ和IL-10较E蛋白组均有明显上升,且差异显著;但攻毒实验发现E蛋白+pCAG-GM组的小鼠全部死亡,生存率为0,而E蛋白组的小鼠保护率为30%。结论 GM-CSF抑制了登革病毒E蛋白的免疫保护作用,其作为蛋白疫苗佐剂仍需慎重。
