Metabolic Profiles of the Kinome Mutants in Fusarium graminearum

  Fusarium head blight, caused by Fusarium graminearum, is one of the most important diseases on wheat and barley.In a previous study, we generated mutants of all the 96 non-essential protein kinase genes and characterized their defects in 17 phenotypes.In this study, we analyzed changes in the metabolite profiles in all these protein kinase mutants by gas chromatography/mass spectrometry (GC/MS) analysis.All the mutants were cultured in minimal medium (MM) for 10 days.Vegetative hyphae were harvested by filtration through Miracloth and ground in liquid nitrogen.The resulting samples were then extracted with methanol and derivalized with N-trimethylsilylimidazole/trimethylchlorosilane (TMSI/TMCS) or methyl chloroformate (MCF).All the GC/MS data were analyzed for common metabolite profiles and clustering for different protein kinase mutants.Interpretation of the resulting heat maps will be presented in connection of the interacting network of these protein kinases based on their orthologs in the budding yeast or their mutant phenotypes in F.graminearum.Interestingly, other than significant reduction in glycerol and arabitol in the mutants of the FgHog1 pathway genes, other two MAP kinase pathways did not cluster in metabolite profiles as expected, suggesting that these linear pathways may have cross-talking with each other or other signaling pathways.Future plans include GC/MS analyses with additional culture media and biological replicates.
