静脉免疫球蛋白(IVIG)在自身免疫病和免疫调节病中的作用模式还很少为人所了解。在体外IVIG对细胞因子释放和细胞因子受体的直接作用以及天然产生的、中和抗细胞因子抗体作用已有报导。作者试图揭示富含IgM和IgA是否会对IVIG的体外免疫调节能力有任何影响.被测制品Pentaglobin分别含有76%IgG和12%IgM及IgA.作者能证明,即使在Pentaglobin浓度为1.0mg/ml时,能明显地抑制混合淋巴细胞反应(MLR)里的同种抗原诱导的增殖.仅含微量IgM和IgA的标准7SIVIG须高到大约10倍浓度时,才能达到相等的同种免疫应答抑制.
The mode of action of intravenous immunoglobulin (IVIG) in autoimmune and immunomodulatory diseases is poorly understood. The direct effect of IVIG on cytokine release and cytokine receptors in vitro and the effect of naturally occurring, neutralizing anti-cytokine antibodies have been reported. The authors sought to reveal whether the presence of IgM and IgA, rich in IgM, would have any effect on IVIG’s ability to modulate immunity in vitro.The Pentaglobin test contained 76% IgG and 12% IgM and IgA respectively.The authors were able to demonstrate that even at a Pentaglobin concentration of 1.0 mg / ml , Significantly inhibited alloantigen-induced proliferation in the mixed lymphocyte reaction (MLR) with standard 7SIVIG containing only trace amounts of IgM and IgA, up to approximately 10-fold concentrations, in order to achieve equivalent suppression of alloimmune responses.
