考察人外周血树突状细胞(DC)递呈可溶性肿瘤抗原激发抗瘤免疫作用。应用改良Steinm an 法分离纯化人外周血DC,同时将MDA、3AO及Wish 三种瘤细胞制成可溶性肿瘤抗原,将二者于体外共育,分析DC表面标记的改变及某些细胞因子产生的水平,并检测其对肿瘤细胞杀伤作用的影响。结果表明,经可溶性肿瘤抗原刺激的DC分泌IL-12及TNFα水平增加,同时高水平表达CD40 、CD80 等表面分子;与对照组相比,显示不同程度的促杀伤作用并有效地促进特异性淋巴细胞增殖。GM-CSF上调DC的上述作用。
Examine human peripheral blood dendritic cells (DCs) presenting soluble tumor antigens to stimulate anti-tumor immunity. Human peripheral blood DCs were isolated and purified by the modified Steinman method. At the same time, MDA, 3AO and Wish tumor cells were made into soluble tumor antigens. The two were co-cultured in vitro to analyze changes in DC surface markers and the production of certain cytokines. Level and detect its effect on tumor cell killing. The results showed that DCs stimulated by soluble tumor antigens secreted increased levels of IL-12 and TNFα and expressed high levels of CD40, CD80, and other surface molecules. Compared with the control group, they showed different degrees of promoting killing and effectively promoted specific lymphoid cells. Cell Proliferation.
