目的:建立同时测定茜草炭中异茜草素、羟基茜草素、1,3,6-三羟基-2-甲基蒽醌、大叶茜草素含量的超高效液相色谱法。方法:采用Acquity BEHC18色谱柱(2.1 mm×50 mm,1.7μm),流动相为甲醇-0.3%甲酸溶液,梯度洗脱,流速0.2 mL.min-1,进样量2μL,柱温30℃,检测波长276 nm。结果:异茜草素、羟基茜草素、1,3,6-三羟基-2-甲基蒽醌、大叶茜草素的质量浓度与峰面积分别在0.69~34.44 mg.L-1(r=0.999 9),0.66~33.2 mg.L-1(r=0.999 7),0.68~34.08 mg.L-1(r=0.999 9),1.07~53.52 mg.L-1(r=0.999 9)呈良好线性;平均回收率分别为96.95%,95.75%,102.5%,96.15%,RSD均小于3%。结论:该方法简便、快速、准确,可作为茜草炭质量控制的一个有效方法。
OBJECTIVE: To establish an ultra performance liquid chromatography (HPLC) method for the simultaneous determination of alismatis, hydroxymeadine, 1,3,6-trihydroxy-2-methylanthraquinone, Methods: The mobile phase consisted of methanol-0.3% formic acid with gradient elution at a flow rate of 0.2 mL.min-1 on an Acquity BEHC18 column (2.1 mm × 50 mm, 1.7 μm) with a sample volume of 2 μL and a column temperature of 30 ° C. Detection wavelength of 276 nm. Results: The concentrations and peak areas of alismatisin, phorboloresol, 1,3,6-trihydroxy-2-methylanthraquinone and magnolia were respectively 0.69-34.44 mg.L-1 (r = 0.999 (R = 0.999 9), 0.66-33.2 mg.L-1 (r = 0.999 7), 0.68-34.08 mg.L-1 (r = 0.999 9) and 1.07-53.52 mg.L- The average recoveries were 96.95%, 95.75%, 102.5% and 96.15%, respectively. The RSDs were less than 3%. Conclusion: The method is simple, rapid and accurate and can be used as an effective method for the quality control of Rubia cordifolia.
