目的:筛选溃疡性结肠炎相关性结肠癌与正常组织表达的差异蛋白。方法:应用蛋白质组学技术,对小鼠结肠癌组织和正常对照组织进行胶内差异双向电泳、(2-D),选择差异表达超过2倍的蛋白点进行MALDI-TOF-MS质谱分析。结果:成功建立结肠癌和对照组织的双向凝胶电泳图谱;对其中差异表达超过2.0倍蛋白质点7个进行了质谱分析和数据库检索鉴定。结论:溃疡性结肠炎相关性结肠癌存在差异表达的蛋白,有可能成为该病潜在的、有应用价值的分子标志。
Objective: To screen differential proteins expressed in ulcerative colitis-associated colon and normal tissues. METHODS: Proteomics technique was used to perform two-dimensional gel electrophoresis (2-D) on mouse colon cancer tissues and normal control tissues. Two-fold differentially expressed protein spots were selected for MALDI-TOF-MS mass spectrometry analysis. RESULTS: Two-dimensional gel electrophoresis profiles of colon cancer and control tissues were successfully established, and 7 of them were differentially expressed at more than 2.0 times of protein spots for mass spectrometry analysis and database search and identification. Conclusion: There are differentially expressed proteins in ulcerative colitis-associated colon cancer, which may be potential and useful molecular markers of the disease.
