目的:建立高效液相色谱法测定苦碟子中木犀草素-7-O-β-D-葡萄糖醛酸苷(L7G)含量,为苦碟子质量标准的制定提供科学依据。方法:采用Luna-C18色谱柱(250mm×4.6mm,5μm),乙腈-0.4%磷酸水溶液(18∶82)为流动相,流速为0.9mL·min-1,检测波长348nm,柱温40℃。结果:以高效液相色谱法测定L7G的回归方程为Y=3.8956+1258.5X(r=0.9999);平均回收率为99.87%,RSD=0.21%;按本文方法对18批样品进行测定,其含量范围为0.12%~0.41%。结论:本方法简便、准确,重复性好,可用来制定L7G的可控质量标准。
OBJECTIVE: To establish a HPLC method for the determination of Luteolin-7-O-β-D-glucuronide (L7G) in Kupin, providing a scientific basis for the establishment of Kudie quality standards. Methods: The mobile phase consisted of Luna-C18 column (250mm × 4.6mm, 5μm) and acetonitrile-0.4% phosphoric acid solution (18:82) with a flow rate of 0.9mL · min-1 and a detection wavelength of 348nm at a column temperature of 40 ℃. Results: The regression equation for the determination of L7G by HPLC was Y = 3.8956 + 1258.5X (r = 0.9999). The average recovery was 99.87% and the RSD was 0.21%. According to the method, 18 samples were determined. The range is from 0.12% to 0.41%. Conclusion: The method is simple, accurate and reproducible. It can be used to develop the controllable quality standard of L7G.
