目的通过构建RNAi表达载体,转染喉癌细胞株Hep-2,观察其对survivin表达的抑制作用。方法构建针对survivin基因的干扰载体,用脂质体转染喉癌细胞Hep-2,分析干扰载体对survivin表达的抑制情况,检测细胞凋亡。结果survivin在喉癌细胞Hep-2中呈高表达,其基因序列与GenBank序列对比一致,干扰载体pGensil/survivin能抑制survivin的表达,抑制率为68%,并诱导了喉癌细胞的凋亡。结论针对survivin的小分子RNA可以有效抑制喉癌细胞Hep-2中survivin蛋白的表达,并促进喉癌细胞凋亡。
Objective To construct a RNAi expression vector and transfect laryngeal carcinoma cell line Hep-2 to observe its inhibitory effect on survivin expression. Methods The interfering vector targeting survivin gene was constructed and transfected into laryngeal carcinoma Hep-2 cells by lipofectamine. The expression of survivin was detected by interfering vector and the apoptosis was detected. Results Survivin was highly expressed in Hep-2 laryngeal carcinoma cells. The gene sequence was consistent with that of GenBank. The expression of survivin was inhibited by pGensil / survivin, and the inhibition rate was 68%. Conclusion Small interfering RNA targeting survivin can effectively inhibit the expression of survivin in Hep-2 laryngeal carcinoma cells and promote the apoptosis of laryngeal carcinoma cells.
