目的 :了解中药鸦胆子粗提成分对正常成纤维细胞 (NF)与瘢痕成纤维细胞 (KF)的影响。方法 :用四氮噻唑蓝 (MTT)法测定经鸦胆子作用后离体培养的NF与KF细胞的存活与增殖能力变化。结果 :两种细胞对鸦胆子反应不同 :1.90 μg/mL可以明显抑制KF细胞增殖 ,2 5 0 μg/mL则可以完全抑制增殖 ;15 .6μg/mL可以明显抑制NF细胞增殖 ,5 0 0 μg/mL才可完全抑制其增殖。NF的IC50值为 42 .2 μg/mL ,KF的IC50 值为 3 7.8μg/mL。结论 :鸦胆子可以抑制NF、KF ,但程度不同 ,抑制KF反应 >NF反应。为瘢痕的临床治疗提供实验依据。改良MTT法 ,有简单、快捷、稳定的优点
Objective : To understand the effects of crude extracts of Brucea javanica on normal fibroblasts (NF) and scar fibroblasts (KF). Methods: The ability of surviving and proliferation of NF and KF cells cultured in vitro by Brucea javanica was determined by tetrazolium blue (MTT) method. RESULTS: The two cells had different responses to Brucea javanica: 1.90 μg/mL could significantly inhibit the proliferation of KF cells, 250 μg/mL could completely inhibit proliferation, and 156 μg/mL could significantly inhibit the proliferation of NF cells, 500 μg. /mL can completely inhibit its proliferation. The IC50 for NF was 42.2 μg/mL and the IC50 for KF was 3 7.8 μg/mL. Conclusion: Brucea javanica can inhibit NF and KF, but the degree is different and inhibit KF response >NF. Provide experimental basis for the clinical treatment of scars. Improved MTT method has advantages of simplicity, speed, and stability
