目的研究大肠癌组织中PCNA的表达意义.方法大肠癌组织63例,其中结肠癌23例,直肠癌40例;男33例,女30例;年龄26岁~76岁.用抗人PCNA单克隆抗体和SP免疫组化技术半定量测定了PCNA在大肠癌组织中的表达,并分析了其与肿瘤分化、侵袭和淋巴结转移间的关系.结果高分化组PCNA阳性Ⅰ~Ⅳ级例数分别为12,2,3,3,中分化组为8,7,4,2,低分化组为5,3,6,8.经Radit统计学分析及显著性检验,低分化大肠癌中PCNA的表达显著高于高、中分化大肠癌(P<005,P<005);其表达与肿瘤的侵袭和淋巴结转移无关(P>005,P>005).结论PCNA的表达可反映大肠癌细胞的增殖活性,是大肠癌增殖生物学行为的有用指标.
Objective To study the expression of PCNA in colorectal cancer. Methods 63 cases of colorectal cancer tissue, including 23 cases of colon cancer, rectal cancer 40 cases; 33 males and 30 females; aged 26 to 76 years old. The expression of PCNA in colorectal cancer tissue was detected semi-quantitatively by anti-human PCNA monoclonal antibody and SP immunohistochemical technique, and the relationship between PCNA and tumor differentiation, invasion and lymph node metastasis was analyzed. Results The number of PCNA positive grades I-IV was 12,2,3,3 in the well-differentiated group, 8,7,4,2 in the moderately differentiated group, and 5, 3, 6 and 8 in the poorly differentiated group. After Radit statistical analysis and significant test, the expression of PCNA in poorly differentiated colorectal cancer was significantly higher than that of high and moderately differentiated colorectal cancer (P<0.05), and its expression was associated with tumor invasion and lymph node metastasis. Irrelevant (P>005, P>005). Conclusion The expression of PCNA can reflect the proliferative activity of colorectal cancer cells, and it is a useful index for the proliferation of colorectal cancer.
