FOXO转录因子是叉头框(forkhead box,FOX)转录因子的一个亚群。因其肿瘤抑制作用而备受关注。FOX家族在人类基因中进化保守,包含从FOXA到FOXR 100多个成员,其序列组成相近。虽然这些成员功能各不相同,但都与Insulin/PI3K/AKT信号通路有关。FOXO转录因子调控的下游靶基因功能广泛,涉及细胞凋亡、细胞周期、氧化应激反应、糖代谢、细胞分化、肌萎缩、稳态、抗衰老、肿瘤抑制等。随着研究的深入,FOXO的信号通路及其调节方式逐渐明了,对揭示肿瘤发生与抑制机制,进而制定临床治疗策略意义重大。
