目的 研究生长激素对全脑缺血 /再灌注后大鼠脑神经细胞的保护作用。方法 SD大鼠 18只 ,雌雄各半 ,随机分 3组 :对照组、CPR 2 4h组及生长激素组 ,每组 6只。采用窒息合并冰氯化钾停跳液致大鼠心跳骤停 -心肺复苏模型 ,停跳 5min后开始CPR ,采用免疫组织细胞化学方法观察各组海马区神经元中bcl- 2和bax的表达情况。结果 生长激素组bcl- 2表达明显较常规复苏组和对照组高 ,而TUNEL阳性细胞少。结论 生长激素可能促进复苏早期神经元表达抗凋亡蛋白bcl- 2 ,抑制神经元的凋亡 ,减轻复苏后海马区神经元的损伤
Objective To study the protective effect of growth hormone on brain cells of rat brain after global cerebral ischemia / reperfusion. Methods Eighteen SD rats were randomly divided into 3 groups: control group, CPR 24h group and growth hormone group, with 6 mice in each group. CPR was induced by asphyxiation combined with ice-cold potassium chloride cardioplegia in rats. CPR was started after 5 minutes of arrest and the expression of bcl-2 and bax in hippocampal neurons of each group was observed by immunohistochemistry . Results The expression of bcl-2 in growth hormone group was significantly higher than that in normal resuscitation group and control group, but less in TUNEL positive cells. CONCLUSIONS: Growth hormone may promote the expression of anti-apoptotic protein bcl-2 in early neurons of resuscitation, inhibit the apoptosis of neurons and alleviate the damage of neurons in hippocampus after resuscitation
