目的:观察地龙水提取物对大鼠大脑中动脉缺血再灌注损伤的影响。方法:大鼠提前14天灌胃地龙水提取物(相当生药量1~4g/kg)后,采用大鼠局灶性脑缺血再灌注损伤模型,观察大鼠行为学变化、大脑中动脉闭塞侧脑梗死面积、脑组织超氧化物歧化酶(S()D)、谷胱甘肽过氧化物酶(GSH-Px)、脂质过氧化物丙二醛(MDA)、白细胞介素(IL)-1β及肿瘤坏死因子(TNF)-α水平的变化。结果:与缺血再灌注损伤组比较,地龙液组脑功能受损程度明显改善,大脑中动脉闭塞侧脑梗死面积明显缩小,脑组织SOD与GSH-Px水平显著升高,MDA、IL-1β、TNF-α水平显著下降且呈剂量依赖关系。结论:地龙液对大鼠大脑缺血再灌注诱导的氧自由基、脂质过氧化、各种炎性因子损伤具有良好的保护作用。
OBJECTIVE: To observe the effect of earthworm water extract on ischemia-reperfusion injury of middle cerebral artery in rats. METHODS: Rats were treated with water extract (amount of crude drug 1~4g/kg) 14 days earlier. The rat model of focal cerebral ischemia-reperfusion injury was used to observe the changes of behavior and the middle cerebral artery. Obstructed lateral cerebral infarct size, brain tissue superoxide dismutase (S(-)D), glutathione peroxidase (GSH-Px), lipid peroxide malondialdehyde (MDA), interleukin ( Changes of IL-1β and tumor necrosis factor (TNF)-α levels. RESULTS: Compared with the ischemia-reperfusion injury group, the degree of brain function impairment was significantly improved in the Dilong Liquid group. The cerebral infarction area of the middle cerebral artery occlusion was significantly reduced, and the SOD and GSH-Px levels in the brain tissue were significantly increased. MDA, IL- The levels of 1β and TNF-α were significantly decreased in a dose-dependent manner. Conclusion: Dilong Liquid has a good protective effect on the injury of oxygen free radicals, lipid peroxidation and various inflammatory factors induced by cerebral ischemia-reperfusion in rats.
