目的观察T3对小鼠糖尿病肾组织中转化生长因子β1(TGF-β1)表达的影响,探讨T3影响糖尿病肾组织纤维化的作用机制。方法 36只小鼠随机挑选12只为正常对照组(C),余24只以注射链尿佐菌素(STZ)方式复制糖尿病小鼠模型,分为糖尿病组(DM)和T3组(T3)。动物饲养6个月后取材肾组织,应用免疫组化技术、体视学方法(检测体密度)和Western blot技术观察各组肾组织中TGF-β1的表达。结果与对照组相比,糖尿病组小鼠TGF-β1的表达范围明显增加,T3组TGF-β1的表达范围明显低于糖尿病组。结论 T3通过降低TGF-β1影响糖尿病肾组织纤维化范围大小和进程。
Objective To observe the effect of T3 on the expression of transforming growth factor β1 (TGF-β1) in the diabetic rat kidney and to explore the mechanism of T3 on the fibrosis in diabetic nephropathy. Methods Thirty - six mice were randomly selected as the normal control group (C), and the other 24 mice were randomly divided into diabetic group (DM) and T3 group (T3) by injection of streptozotocin (STZ) . Six months after the feeding, the kidneys were harvested, and the expression of TGF-β1 in each group was observed by immunohistochemistry, stereology and Western blot. Results Compared with the control group, the expression of TGF-β1 in diabetic mice increased significantly, and the expression of TGF-β1 in T3 group was significantly lower than that in diabetic rats. Conclusion T3 can reduce the extent and process of TGF-β1 in diabetic nephropathy.
