目的评价血管生成抑制剂 TNP-470与碘化油混合栓塞对兔肝癌的抑瘤效果.方法将 VX_2瘤细胞接种于24只新西兰白兔肝左叶,建立兔肝癌模型,随机分4组,每组6只.利用导管经肝动脉分别给碘油、TNP-470,TNP-470+碘油及生理盐水于不同组,1 wk 后观察各组肿瘤体积、肿瘤微血管密度及血清 AST 水平,并观察存活期及组织病理切片.结果 TNP-470+碘油组生长率为(0.62±0.28)与对照组(3.48±1.17)相比差异有显著性(P<0.01),与栓塞组(1.28±0.23)、TNP-470组(1.18±0.26)相比差异有显著性(P<0.05).混合栓塞组微血管密度(MVD 值)(9.98±5.71)与对照组(23.12±9.67)相比差异有显著性(P<0.01).混合栓塞组存活期(53.0±2.0)与对照组(40.5±3.0)相比差异有显著性(P<0.05).混合栓塞组血清 AST 水平治疗前后变化与其他处理相比差异无显著性(P>0.05),与对照组相比差异有显著性(P<0.05).混合栓塞组病理 HE 染色切片示:有早期坏死表现.结论 TNP-470与碘油混合栓塞组可大大降低肿瘤生长率,减小微血管密度,延长存活期,并对肝功损害是可逆的,其抑瘤效果更强.
Objective To evaluate the antitumor effect of mixed embolization of angiogenesis inhibitor TNP-470 and iodized oil on rabbit hepatocarcinoma.Methods VX_2 tumor cells were inoculated into the left lobe of the liver of 24 New Zealand white rabbits to establish a model of rabbit liver cancer and were randomly divided into 4 groups Group 6. The tumor volume, tumor microvessel density and serum AST levels of each group were observed by different groups after administration of lipiodol, TNP-470, TNP-470 + lipiodol and normal saline via hepatic artery in different groups for 1 week Survival time and histopathological sections.Results The growth rate of TNP-470 + lipiodol group was (0.62 ± 0.28) compared with that of control group (3.48 ± 1.17) (P <0.01) ), TNP-470 group (1.18 ± 0.26) (P <0.05), there was significant difference between the MVD value of the mixed embolization group (9.98 ± 5.71) and the control group (23.12 ± 9.67) (P <0.01). The survival time of the mixed embolization group (53.0 ± 2.0) was significantly different from that of the control group (40.5 ± 3.0) (P <0.05) (P> 0.05), but the difference was significant compared with the control group (P <0.05) .Hematoxylin and Eosin Conclusions The combination of TNP-470 and lipiodol embolization can significantly reduce tumor growth rate, reduce microvessel density, prolong survival, and is reversible to liver damage, and its anti-tumor effect is stronger.
