目的 研究青春期小鼠Leydig细胞中转醛酶活性、蛋白表达水平与睾酮生成的相关性。 方法 原代培养Leydig细胞 ,加入HCG刺激 ,酶联免疫法测定培养 2、8小时细胞上清睾酮浓度 ,同时测定转醛酶活性 ,Westernblot检测转醛酶蛋白表达水平。结果 (1)刺激组Leydig细胞上清睾酮浓度明显高于对照组 ,组间存在显著性差异 (2小时P <0 .0 1,8小时P <0 .0 0 1)。 (2 )刺激组Leydig细胞转醛酶活性明显高于对照组 ,组间存在显著性差异(2小时P <0 .0 5 ,8小时P <0 .0 0 1)。 (3)Leydig细胞刺激组和对照组间转醛酶蛋白表达水平无显著差异 (P >0 .0 5 )。结论 HCG促进小鼠Leydig细胞生成睾酮的同时 ,转醛酶活性同步提高 ,证实转醛酶活性与小鼠Leydig细胞合成睾酮密切相关
Objective To study the correlation between transaldolase activity and protein expression in adolescent Leydig cells and testosterone production. Methods Primary cultured Leydig cells were stimulated with HCG. The serum concentration of testosterone was measured by enzyme-linked immunosorbent assay (ELISA) for 2 and 8 hours. The transaldolase activity was measured and the transaldolase protein expression was detected by Western blot. Results (1) The concentration of testosterone in the supernatant of Leydig cells in the stimulation group was significantly higher than that in the control group, and there was a significant difference between the two groups (P <0.01 for 2 hours and P <0.01 for 2 hours). (2) The transaldolase activity of Leydig cells in stimulation group was significantly higher than that in control group, there was significant difference between two groups (P <0.05 for 2 hours, P <0.01 for 8 hours). (3) There was no significant difference in transaldolase protein expression between Leydig cell stimulation group and control group (P> 0.05). Conclusions HCG can promote the production of testosterone in Leydig cells, meanwhile, the activity of transaldolase increases synchronously, which confirms that the activity of transaldolase is closely related to the synthesis of testosterone in Leydig cells
