目的:探讨人参皂苷Rg1经PEG修饰后在小鼠的组织靶向分布情况。方法:取SD小鼠随机分为2组,分别给予尾静脉注射Rg1和PEG-Rg1,于不同时间点取血并取各组织脏器,采用UPLC对样品中指标成分人参皂苷Rg1进行含量测定,计算Rg1在各组织分布的靶向系数,观察比较2组在小鼠体内的组织靶向分布情况。结果:人参皂苷Rg1组在小鼠体内各组织分布的AUC大小顺序为肝、肾、肺、心、脾,肝靶向系数为2.01;PEG修饰组在小鼠体内各组织的AUC大小顺序为肾、肝、肺、心、脾,肝靶向系数为9.21。结论:PEG修饰后,Rg1对肝组织的靶向选择性增强,同时对肾、肺组织的靶向选择性也得到增强。
Objective: To investigate the tissue-targeted distribution of ginsenoside Rg1 in mice after PEG modification. Methods: SD mice were randomly divided into two groups. Rg1 and PEG-Rg1 were injected into the caudal vein respectively. Blood samples were obtained from different tissues at different time points. The content of ginsenoside Rg1 in the sample was determined by UPLC. The targeting coefficient of Rg1 distribution in each tissue was calculated, and the tissue targeting distribution of the two groups in mice was observed and compared. Results: The AUC order of the distribution of ginsenoside Rg1 in different tissues of mice was 2.01, and the AUC order of liver, kidney, lung, heart, spleen and liver was 2.01. The order of AUC of PEG modified group in mice was kidney , Liver, lung, heart, spleen, liver targeting coefficient of 9.21. Conclusion: After modified with PEG, targeting selectivity of Rg1 to liver tissue is enhanced, and targeted selectivity to kidney and lung tissue is also enhanced.
