目的 探讨caspase 3、bcl 2蛋白在非霍奇金淋巴瘤 (NHL)发生、发展中的可能作用及相互关系。方法 应用TdT介导的dUTP缺口末端标记 (TUNEL)技术和免疫组织化学链霉素抗生物素 过氧化酶 (SP)法 ,检测 5例反应性增生淋巴组织和 119例NHL组织中的细胞凋亡和增殖细胞核抗原(PCNA)、caspase 3、bcl 2蛋白的表达水平。结果 caspase 3和bcl 2在 119例NHL中表达的阳性率分别为 86 6 %(10 3例 )和 5 3.8%(6 4例 )。二者在良、恶性淋巴组织中的表达方式相反 :在反应性增生淋巴滤泡中心caspase 3高表达而bcl 2阴性表达 ,滤泡套区caspase 3阴性表达而bcl 2高表达 ;在肿瘤性滤泡中心bcl 2常高表达而caspase 3常表达减弱或不表达 ;在NHL中二者的表达与肿瘤恶性度呈相反关系 ,高度恶性组中caspase 3的表达 (4 4.4%)高于低度恶性组 (2 3.7%,P <0 .0 1) ,而B细胞性淋巴瘤中bcl 2的表达 (4 2 .6 %)低于低度恶性组 (75 .5 %,P <0 .0 1) ;caspase 3的表达与凋亡指数呈正相关(r=0 .5 12 ,P <0 .0 1) ) ,bcl 2的表达与凋亡指数呈负相关 (r =- 0 .436 ,P <0 .0 1)。此外 ,NHL的凋亡指数与增殖指数呈显著正相关 (r =0 .710 ,P <0 .0 1)。结论 caspase 3可能参与了NHL的凋亡调节机制。caspase 3和bcl 2蛋白在良、恶性淋巴组?
Objective To investigate the possible roles and relationships of caspase 3 and bcl 2 proteins in the occurrence and development of non-Hodgkin’s lymphoma (NHL). Methods TdT-mediated dUTP nick end labeling (TUNEL) technique and immunohistochemical streptavidin peroxidase (SP) method were used to detect apoptosis in 5 cases of reactive hyperplasia and 119 NHL tissues. Expression levels of proliferating cell nuclear antigen (PCNA), caspase 3, and bcl 2 proteins. Results The positive rates of caspase 3 and bcl 2 expression in 119 cases of NHL were 86.6% (10 3 cases) and 5 3.8% (64 cases), respectively. The expression patterns of the two were opposite in benign and malignant lymphoid tissues: in the reactive hyperplastic follicle center, caspase 3 was highly expressed and bcl 2 was negatively expressed, follicular caspase 3 was negatively expressed and bcl 2 was highly expressed; The expression of bcl 2 in the vesicle center was often high and caspase 3 was usually weakly or not expressed. The expression of both caspase 3 in the NHL was inversely related to the degree of malignancy of the tumor. The expression of caspase 3 in the high-grade group (4 4.4%) was higher than low grade. In group B (2 3.7%, P <0.01), the expression of bcl 2 in B-cell lymphoma (42.6%) was lower than that in low-grade group (75.5%, P <0.01). The expression of caspase 3 was positively correlated with the apoptotic index (r=0.51, P<0.01). The expression of bcl-2 was negatively correlated with the apoptotic index (r =- 0.436 ,P < 0 .0 1). In addition, there was a significant positive correlation between the apoptosis index and proliferation index of NHL (r = 0.710, P < 0.01). Conclusion caspase 3 may be involved in the regulation of apoptosis in NHL. Are caspase 3 and bcl 2 proteins in the benign and malignant lymph nodes?
