目的研究葛根芩连汤含药血清体外抑制轮状病毒(RV)的作用及其机制。方法采用观察CPE及MTT法,从抑制病毒生物合成、直接灭活病毒、抗病毒吸附三方面,测定葛根芩连汤含药血清对RV感染MA104细胞病变的作用。结果抑制病毒生物合成、直接灭活病毒方面,与相应浓度空白血清比较,各浓度葛根芩连汤含药血清与西药组含药血清使RV感染MA104细胞OD值明显升高(P<0.01);在抗病毒吸附方面,各浓度葛根芩连汤含药血清作用的细胞OD值明显高于同浓度空白组和西药组(P<0.01)。结论葛根芩连汤主要通过直接灭活轮状病毒以及抑制轮状病毒生物合成而发挥其抗病毒作用。
Objective To study the inhibitory effect of Gegen Qinlian Decoction-containing serum on rotavirus (RV) and its mechanism. Methods The effects of Gegen Qinlian Decoction-containing serum on the pathology of RV-infected MA104 cells were determined by observing the CPE and MTT methods from the aspects of inhibiting the virus biosynthesis, directly inactivating the virus and antivirus adsorption. Results Compared with the corresponding blank serum, the OD value of RV infected MA104 cells was significantly increased (P <0.01) with the drug - containing serum of Gegen Qinlian Decoction and that of western medicine group. In terms of antiviral adsorption, the OD value of Gegen Qinlian Decoction drug-containing serum was significantly higher than that of blank control group and western medicine group (P <0.01). Conclusion Gegen Qinlian Decoction can exert its antiviral effect mainly by direct inactivation of rotavirus and inhibition of rotavirus biosynthesis.
