目的:研究整合素αvβ6亚基αv及β6对高转移卵巢癌细胞系HO-8910PM增殖及侵袭的影响。方法:流式细胞仪检测HO-8910PM细胞αv及β6阳性表达率,MTT实验观察抗αv及抗β6单抗对HO-8910PM细胞增殖的影响,细胞侵袭实验观察抗αv及β6单抗对该细胞系侵袭的影响。结果:HO-8910PM细胞胞质内表达αvβ6,但表达率均不高,αv阳性表达率为25.7%,β6为28.3%;MTT实验显示,αv单抗可明显抑制HO-8910PM细胞增殖(P<0.05),β6单抗抑制肿瘤增殖作用不明显。细胞侵袭实验显示,β6单抗能明显抑制HO-8910PM细胞对人工基底膜胶的侵袭穿透能力(P<0.05)。结论:整合素αvβ6的αv亚基主要参与卵巢癌肿瘤细胞的增殖过程,β6亚基更多参与卵巢癌肿瘤细胞的侵袭过程。
Objective: To investigate the effects of integrin αvβ6 subunit αv and β6 on the proliferation and invasion of highly metastatic ovarian cancer cell line HO-8910PM. Methods: The positive rate of αv and β6 in HO-8910PM cells was detected by flow cytometry. The effect of anti-αv and anti-β6 monoclonal antibody on the proliferation of HO-8910PM cells was observed by MTT assay. The impact of invasion. Results: The expression of αvβ6 in the cytoplasm of HO-8910PM cells was not significant, the positive expression rate of αv was 25.7% and that of β6 was 28.3%. MTT assay showed that αv monoclonal antibody could significantly inhibit the proliferation of HO-8910PM cells (P < 0.05), β6 monoclonal antibody inhibition of tumor proliferation is not obvious. Cell invasion assay showed that β6 monoclonal antibody could significantly inhibit the invasion and penetration of HO-8910PM cells (P <0.05). CONCLUSION: The αv subunit of integrin αvβ6 is mainly involved in the proliferation of ovarian cancer cells. The β6 subunit is more involved in the invasion of ovarian cancer cells.
