采用细胞培养法 ,免疫组化实验技术和流式细胞仪等项技术 ,研究了bcl 2基因反义核酸和砷剂单独及联合应用对K5 6 2细胞的生物效应 ,DNA及bcl 2蛋白的变化。结果显示 ,As2 O3(2 .0 μmol L) +bcl 2ASODN(10 .0 μmol L)联合作用比单用As2 O3 (2 .0 μmol L)或bcl 2ASODN(10 .0 μmol L)显著增强诱导细胞凋亡的作用 (P <0 .0 1) ;流式细胞仪检测显示 ,联合作用引起的bcl 2蛋白表达亦明显减少 (P <0 .1)。结论表明 ,bcl 2基因反义核酸能明显提高和显著增强K5 6 2白血病细胞对As2 O3 药物敏感性
Cell culture, immunohistochemistry and flow cytometry were used to study the biological effects of bcl 2 antisense nucleic acid and arsenic alone and in combination on K562 cells, and changes in DNA and bcl 2 proteins. . The results showed that the combined effect of As2O3 (2.0 μmol L) + bcl2ASODN (10.0 μmol L) significantly enhanced the induction of cells than either As2O3 alone (2.0 μmol L) or bcl 2ASODN (10.0 μmol L). The effect of apoptosis (P <0.01) was detected by flow cytometry, and the expression of bcl-2 protein was also significantly reduced (P <0. 1). The results showed that bcl 2 antisense oligodeoxynucleotide can significantly enhance and significantly enhance the sensitivity of K562 cells to As2O3 drugs.
