近来的研究表明小RNA具有调控干细胞自我更新和定向分化的功能,间充质干细胞能分化为三胚层的一些细胞。但至今尚不清楚间充质干细胞是否能转化为造血干细胞,也不清楚小RNA能否调控此过程的发生。由于对这些问题的深入和全面地研究,有助于开辟新的造血途径,了解干细胞定向分化的分子机制,也有助于解决造血干细胞临床应用中的许多实际问题。在国家“863”专题的支持下,我们使用易于获得的人体脂肪间充质干细胞为研究对象(不需要配型、不需要干细胞库,无免疫排斥,可大量扩增),通过生物信息学和小RNA芯片筛选和鉴定一组新的人内源性小RNA,
Recent studies have shown that small RNAs have the ability to regulate stem cell self-renewal and directed differentiation, and mesenchymal stem cells can differentiate into some cells of the three germ layers. However, it is still not clear whether mesenchymal stem cells can be transformed into hematopoietic stem cells. It is not clear whether small RNA can regulate the occurrence of this process. Because of these issues in-depth and comprehensive study, help to open up new hematopoietic pathways, understand the molecular mechanisms of stem cell differentiation, but also help solve many practical problems in the clinical application of hematopoietic stem cells.
