目的 观察血小板激活因子 (PAF)对血小板聚集性、5 HT释放、胞浆内游离钙离子浓度的影响及红花总黄色素的抑制作用。方法 红花水煎液经硅胶吸附法除杂质得到红花总黄色素 ,以比浊法及邻苯二甲醛 (OPT)荧光分光光度法观察其对PAF诱导家兔洗涤血小板 (WRP)聚集及 5 HT释放的影响。以Fura 2荧光分光光度法测定血小板内游离钙离子浓度。结果 红花总黄色素抑制PAF诱发的WRP聚集、5 HT释放作用呈明显的量效关系。PAF 2 .0× 10 - 9mol·L- 1 时 ,0 .2 1,0 .42 ,0 .85 ,1.6 9g·L- 1 红花总黄色素血小板聚集抑制率依次为 2 8.0 % ,32 .9% ,6 0 .0 % ,79.4% ;5 HT释放抑制率分别为 4.70 % ,12 .9% ,32 .9% ,5 8.4% ;同时 4.1,6 .5 ,10 .3 ,16 .7,2 6 .7mg·ml- 1 红花总黄色素剂量依赖地抑制 8× 10 - 1 0 mol·L- 1 PAF引起的血小板内游离钙增高。结论 红花总黄色素可抑制PAF诱导的血小板聚集、5 HT释放及血小板内游离钙增加
Objective To observe the effects of platelet activating factor (PAF) on platelet aggregation, 5 HT release, intracellular free calcium ion concentration and inhibition of total safflower yellow pigmentation. Methods Saffron decoction was decontaminated by silica gel to obtain safflower total yellow pigment. TPR and OPT fluorescence spectrophotometry were used to observe PAF-induced WRP aggregation in rabbits and 5 Effect of HT release. Platelet intracellular free calcium ion concentration was measured by Fura 2 fluorescence spectrophotometry. Results The total yellow pigment of safflower inhibited the WRP accumulation and 5 HT release induced by PAF in a dose-dependent manner. When PAF 2 .0×10 −9 mol·L-1, the inhibitory rates of platelet aggregation were 0.28%, 0.25%, 1.65% and 1.69%, respectively. 9%, 60%, 79.4%; 5 HT release inhibition rates were 4.70 %, 12.9%, 32.9%, 58.4 %; meanwhile 4.1, 6 .5, 10 .3, 16. 7 26.7 mg·ml - 1 safflower total yellow inhibited the increase of platelet free calcium induced by 8 × 10 - 10 mol·L - 1 PAF dose-dependently. Conclusion Safflor yellow can inhibit PAF-induced platelet aggregation, 5 HT release, and platelet free calcium increase
