用成年雄性Wistar大鼠,分为溃疡组和对照组。用免疫电镜包埋后免疫金间接法标记ACTH细胞,研究胃溃疡自愈过程中垂体远侧部ACTH细胞的超微结构变化。结果显示,在实验性胃溃疡第4d,一些ACTH细胞含少量很小的分泌颗粒,沿质膜分布。有较多的ACTH细胞胞质出现许多空泡。溃疡第6d,ACTH细胞的高尔基复合体发达,其扁囊膨大部含中等电子密度的内容物,其内可见金颗粒标记。溃疡6~10d,较多的ACTH细胞含许多分泌颗粒。溃疡第28d,ACTH细胞的超微结构基本恢复正常。本研究结果表明,在大鼠实验性胃溃疡自愈期间,垂体远侧部ACTH细胞的超微结构显示明显的变化。这些变化主要反映了ACTH细胞合成、贮存及释放激素的活动增强,提示垂体远侧部ACTH细胞参与了溃疡自愈过程的调节。
Adult male Wistar rats were divided into ulcer group and control group. Immunocytochemistry was used to label the ACTH cells by immunohistochemistry to study the ultrastructural changes of ACTH cells in distal part of pituitary during the process of gastric ulcer healing. The results showed that on the 4th day of experimental gastric ulcer, some ACTH cells contained a few small secretory granules distributed along the plasma membrane. There are many vacuoles in the cytoplasm of ACTH cells. On the 6th day of ulcer, the Golgi complex of ACTH cells developed, and the content of medium electron density in the expanded part of the cyst was visible. Ulcers 6 ~ 10d, more ACTH cells contain many secretory granules. On the 28th day of ulcer, the ultrastructure of ACTH cells returned to normal. The results of this study show that during the healing of experimental gastric ulcer in rats, the ultrastructure of ACTH cells in the distal pituitary showed a significant change. These changes mainly reflect the activity of synthesis, storage and release of hormones in ACTH cells, suggesting that ACTH cells in the distal part of pituitary are involved in the regulation of ulcer healing process.
