目的探索miR-204在衰老心肌细胞缺氧后处理中的作用。方法经D-半乳糖诱导心肌细胞衰老后,随机分为对照组(ONC组)、缺氧复氧组(OAR组)、后处理组(OIPO组)。采用ELISA法检测培养基中c Tn-Ι的含量;微板法检测LDH的含量;XTT法检测细胞活性;荧光定量PCR法检测miR-204的mRNA表达,并确定miR-204与c Tn-Ι、LDH的相关性。结果与ONC组相比,OAR、OIPO组c Tn-Ι、LDH的含量均增加(P<0.05,P<0.01);与OAR组相比,OIPO组c Tn-Ι、LDH的含量无显著性改变。与ONC组相比,OAR、OIPO组miR-204的表达均降低(P<0.01);相关性分析显示miR-204与c Tn-Ι、LDH呈现负相关。结论缺氧后处理对缺氧复氧所致的衰老心肌细胞损伤的保护作用减弱与miR-204有关。
Objective To explore the role of miR-204 in post-hypoxic treatment of aged cardiomyocytes. Methods After myocardial cell senescence induced by D-galactose, the rats were randomly divided into the control group (ONC group), the hypoxia-reoxygenation group (OAR group) and the post-treatment group (OIPO group). The content of cTn-1 in culture medium was detected by ELISA, the content of LDH was detected by microplate method, the activity of cell was detected by XTT method, the mRNA expression of miR-204 was detected by fluorescence quantitative PCR, and the expression of miR-204 and cTn-1 , LDH correlation. Results Compared with ONC group, the contents of cTn-1 and LDH in OIPO group were increased (P <0.05, P <0.01). Compared with OAR group, the content of cTn-1 and LDH in OIPO group was not significant change. Compared with ONC group, the expression of miR-204 in both OAR and OIPO groups was decreased (P <0.01). Correlation analysis showed that miR-204 was negatively correlated with cTn-1 and LDH. Conclusions The protective effect of hypoxia postconditioning on the injury of aged cardiomyocytes induced by hypoxia-reoxygenation is related to miR-204.
