目的检测转录因子Fli-1蛋白在外周血T细胞与B细胞上的表达水平,分析其表达水平与系统性红斑狼疮(systemic lupus erythematosus,SLE)患者疾病活动度、实验室指标及临床表现之间的关系。方法选取来自安徽医科大学附属省立医院的28例SLE患者作为病例组,选择25例健康志愿者作为正常对照。利用流式细胞仪(flow cytometry,FCM)检测外周血单核细胞(peripheral blood mononuclear cells,PBMCs)T细胞和B细胞中Fli-1蛋白表达情况。结果 SLE患者外周血T细胞上Fli-1平均荧光强度(mean fluorescence intensity,MFI)高于对照组,差异有统计学意义(t=3.28,P=0.003),而SLE病例与对照组间,Fli-1在外周血B细胞上的表达差异无统计学意义(Z=-0.87,P=0.383)。SLE病人外周血T/B细胞上Fli-1的表达水平与糖皮质激素的平均用量间均呈正相关(均有P<0.05),但并未发现Fli-1在T细胞和B细胞上的表达水平与SLE疾病活动指数间的关联。对多项临床实验室检测指标与Fli-1的关联分析结果显示,低补体的SLE患者,其Fli-1在外周血T细胞与B细胞上的表达水平均高于无低补体表现的SLE患者(均有P<0.05)。结论 SLE病人外周血T细胞上Fli-1表达水平增加,提示Fli-1可能参与了SLE的发病,其具体作用机制仍有待进一步研究。
Objective To detect the expression of Fli-1 protein on peripheral blood T cells and B cells, and to analyze the relationship between the expression of Fli-1 protein and the disease activity, laboratory parameters and clinical manifestations in patients with systemic lupus erythematosus (SLE) Relationship. Results The mean Fli-1 mean fluorescence intensity (MFI) of peripheral blood T cells in SLE patients was significantly higher than that in controls (t = 3.28, P = 0.003) -1 in peripheral blood B cells showed no significant difference (Z = -0.87, P = 0.383). The results of multiple clinical laboratory tests and Fli-1 analysis showed that the expression level of Fli-1 in peripheral blood T cells and B cells in patients with SLE with low complement was higher than those with SLE without low complement (All P <0.05). Conclusion The expression of Fli-1 on peripheral blood T cells in patients with SLE is increased, suggesting that Fli-1 may be involved in the pathogenesis of SLE. The specific mechanism remains to be further studied.
