目的:使用响应面优化法,确定从西洋参茎叶中提取多糖的最佳提取工艺,并且探讨西洋参茎叶多糖的抗病毒活性。方法:设计单因素实验,并在单因素实验的基础上进行响应面实验设计,确定最佳提取工艺。提取的多糖样品,以利巴韦林为对照,RSV-Hep2细胞体外病毒感染模型进行体外抗RSV作用的对比实验。结果:经优化实验得到的最佳工艺为:提取时间为90 min,提取次数为2次,料液比为1:35,最佳优化结果所得多糖的抗病毒TI指数为32.57。结论:在使用响应面软件优化下,确定了最佳提取工艺,并且发现得到的茎叶多糖具有抗病毒作用,对西洋参茎叶多糖的进一步研究提供依据。
OBJECTIVE: To determine the optimal extraction process of polysaccharides from the stems and leaves of American ginseng by response surface methodology, and to study the anti-viral activity of polysaccharides from the stems and leaves of American ginseng. Methods: The single factor experiment was designed. Based on the single factor experiment, the experiment of response surface was designed to determine the best extraction technology. Extracted samples of polysaccharides, ribavirin as a control, RSV-Hep2 cells in vitro virus infection model in vitro anti-RSV effect of the comparative experiment. Results: The optimum process was optimized as follows: the extraction time was 90 min, the number of extraction was 2, the ratio of solid to liquid was 1:35, and the best TI was 32.57. Conclusion: Under the software optimization of response surface, the best extraction technology was determined, and the polysaccharides of stem and leaf obtained were found to have antiviral activity, which provided the basis for further study of polysaccharide from stem and leaf of American ginseng.
