目的采用两种吸收模型考察冰片对绿原酸口服吸收的促进作用并对模型间的研究结果进行相关性评价。方法首先采用大鼠原位灌注模型考察冰片对绿原酸的肠道吸收是否具有促进作用,然后利用整体动物模型进行验证,并对两种模型得到的吸收参数进行相关性评价。结果大鼠原位灌注模型中,合用冰片后,绿原酸吸收速率常数为0.083 4 h-1,比单用绿原酸组的吸收速率常数增加1.58倍;整体动物实验表明,绿原酸灌胃给药后,体内过程符合二室模型,合用0.04%冰片后,最大血药浓度略有增加,相对生物利用度达到116.02%;相关性研究表明,两种模型的研究结果呈正相关,r=0.941,研究结果可靠。结论冰片对绿原酸的口服吸收具有促进作用。
Objective To investigate the effects of borneol on the oral absorption of chlorogenic acid by two absorption models and to evaluate the correlation between the results of the models. Methods The rat model of in situ perfusion was used to investigate whether borneol could promote the intestinal absorption of chlorogenic acid. Then, the animal model was used to verify and the correlation between the absorption parameters obtained from the two models was evaluated. Results In rat model of in situ perfusion, chlorogenic acid absorption rate constant was 0.083 4 h-1, which was 1.58 times higher than that of chlorogenic acid alone group. The overall animal experiments showed that chlorogenic acid irrigation After gastric administration, the in vivo process accorded with two-compartment model. The maximal plasma concentration increased slightly with the increase of 0.04% borneol and the relative bioavailability reached 116.02%. The correlation study showed that there was a positive correlation between the two models, r = 0.941, the research result is reliable. Conclusion Borneol has a positive effect on the oral absorption of chlorogenic acid.
