目的:探讨增殖诱导配体APRIL,X染色体连锁凋亡抑制蛋白XIAP及半胱氨酸天冬氨酸蛋白酶caspase-9在人肝癌组织中的表达及意义。方法:采用实时荧光定量逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)方法,分别检测62例肝癌患者的肝癌组织、癌旁组织及30例正常人的肝脏组织中APRIL,XIAP和caspase-9的表达。对比分析APRIL,XIAP和caspase-9的表达与肝癌临床病理参数的关系。结果:在肝癌组织中,APRIL和XIAP的表达明显高于其在癌旁组织及正常肝脏组织中的表达(P<0.05);caspase-9的表达则明显低于癌旁组织以及正常肝脏组织(P<0.05);在癌旁组织和正常肝脏组织中,APRIL,XIAP和caspase-9的表达无显著差异(P>0.05)。APRIL和XIAP在肝癌组织中的表达与甲胎蛋白、肿瘤组织学分级、临床病理TNM分级有关(P<0.05)。结论:APRIL,XIAP和caspase-9在肝癌组织中的表达与肝癌临床病理特征密切相关,不仅是检测肝癌的重要生物学标志,也是潜在的肝癌治疗靶点。
OBJECTIVE: To investigate the expression and significance of proliferation-inducing ligand APRIL, X-linked inhibitor of apoptosis protein XIAP and caspase-9 in human hepatocellular carcinoma. Methods: APRIL was detected by RT-PCR in liver tissues, adjacent tissues and 30 normal liver tissues of 62 HCC patients respectively. , XIAP and caspase-9 expression. The relationship between the expression of APRIL, XIAP and caspase-9 and clinicopathological parameters of hepatocellular carcinoma was analyzed. Results: The expression of APRIL and XIAP in HCC tissues was significantly higher than that in paracancer tissues and normal liver tissues (P <0.05), while the expression of caspase-9 was significantly lower than that in paracancerous tissues and normal liver tissues P <0.05). There was no significant difference in the expression of APRIL, XIAP and caspase-9 in adjacent tissues and normal liver tissues (P> 0.05). The expression of APRIL and XIAP in hepatocellular carcinoma was correlated with the level of α-fetoprotein, tumor histological grade and clinicopathologic TNM grade (P <0.05). Conclusion: The expression of APRIL, XIAP and caspase-9 in hepatocellular carcinoma is closely related to the clinicopathological features of hepatocellular carcinoma. It is not only an important biological marker for detecting hepatocellular carcinoma, but also a potential therapeutic target for hepatocellular carcinoma.
