目的研究伊班膦酸钠(IBN)对体外培养的人肺巨细胞癌高转移株PLA-801D细胞生长增殖的影响及其潜在的作用机制。方法采用MTT法检测不同浓度IBN、及相同浓度不同作用时间的IBN对PLA-801D细胞系生长增殖的影响,应用流式细胞仪检测不同浓度IBN作用下,PLA-801D细胞周期的变化。结果在10~100μg/ml的浓度范围内,IBN对PLA-801D细胞生长增殖均有抑制作用(P<0.01),抑制效果与IBN的药物浓度和作用时间呈正相关(r药物浓度=0.987,r作用时间=0.985);IBN可使细胞明显阻滞于S期,G2/M期细胞比例则明显减少。结论在10~100μg/ml的浓度范围内,IBN对体外生长的PLA-801D细胞的抑制作用呈剂量、时间依赖性,其机制可能与其将细胞阻滞在DNA合成期(S期),阻碍肿瘤细胞分裂增殖有关。
Objective To investigate the effect of ibandronate (IBN) on the growth and proliferation of human lung giant cell carcinoma high metastasis PLA-801D cells in vitro and its potential mechanism. Methods The effects of different concentrations of IBN and different concentrations of IBN on proliferation and proliferation of PLA-801D cell lines were detected by MTT assay. Flow cytometry was used to detect the changes of PLA-801D cell cycle under different concentrations of IBN. Results In the concentration range of 10-100 μg/ml, IBN inhibited the growth and proliferation of PLA-801D cells (P<0.01), and the inhibitory effect was positively correlated with the drug concentration and duration of IBN (r drug concentration=0.987, r). The time of action was = 0.9985); IBN significantly blocked cells in S phase, and the ratio of cells in G2/M phase decreased significantly. Conclusion In the concentration range of 10-100 μg/ml, the inhibitory effect of IBN on PLA-801D cells grown in vitro was dose- and time-dependent, and its mechanism may be related to its arrest of cells in the DNA synthesis phase (S phase), blocking tumors. Cell division is related to proliferation.
