[目的]探讨尿毒灵(由黄芪、当归、陈皮、法夏、益母草、大黄、蚕砂等组成)对慢性肾功能衰竭(chronic、renalfailure,CRF)大鼠肾功能的影响及其与肾脏转化生长因子(TGF)β_1表达的关系。[方法]采用5/6肾大部切除法致CRF模型大鼠,将30只存活的CRF模型大鼠随机分为模型组、治疗组、对照组3组,另以8只假手术大鼠作假手术组,检测大鼠生化、代谢、病理,以免疫组化法检测TGFβ_1在肾脏的表达,以计算机图象分析法进行半定量比较。[结果]模型组见肾功能减退、电解质紊乱、血色素下降及CRF病理表现;两用药组可使肾衰改变减轻,但组间无显著性差异(P>0.05)。两用药组能显著调低CRF大鼠肾小球硬化指数(GSI)和 TGFβ_1的表达(与模型组比较,均P<0.01),但两治疗组组间比较无显著性差异。[结论]尿毒灵可明显改善CRF大鼠肾功能,同时有使TGFβ_1表达下调的作用,提示尿毒灵可能是通过这一下调作用而改善肾功能。
[Objective] To investigate the effect of uremic ray (composed of astragalus, angelica, tangerine peel, facia, motherwort, rhubarb, silkworm etc.) on renal function in rats with chronic renal failure (CRF) and its relationship with renal growth The relationship between factor (TGF) β_1 expression. [Methods] CRF model rats were induced by 5/6 resection of the kidneys, and 30 surviving CRF rats were randomly divided into 3 groups: model group, treatment group and control group. Another 8 sham-operated rats were falsified. In the operation group, biochemistry, metabolism and pathology of the rats were measured. The expression of TGFβ_1 in the kidney was detected by immunohistochemistry and semi-quantitatively compared by computer image analysis. [Results] The model group showed decreased renal function, electrolyte disturbance, decreased hemoglobin and pathological manifestations of CRF. The dual drug group could reduce the change of renal failure, but there was no significant difference between the two groups (P>0.05). The dual-agent group significantly reduced the glomerular sclerosis index (GSI) and TGFβ_1 expression in CRF rats (P<0.01 compared with the model group), but there was no significant difference between the two treatment groups. [Conclusion] Ureomycin can significantly improve the renal function of CRF rats, and at the same time, it can down-regulate the expression of TGFβ_1, suggesting that uremic spirit may improve renal function through this down-regulation.
