针对中国首例实验室确诊输入性MERS-CoV感染病例,采用鼻咽拭子样品进行核酸提取、基因扩增与测序,获得MERS-CoV_ChinaGD01结构蛋白与附属蛋白编码基因,包括S基因、E基因、M基因、N基因、ORF3、ORF4a、ORF4b、ORF5和ORF8b基因序列。根据S基因进化分析发现中国输入性病例中MERS-CoV虽有少数位点变异,但仍与近年沙特流行株相近,属于MERS-CoV亚型5,N、E、M等结构基因核苷酸变异较少。附属蛋白进化分析表明:ORF3、ORF4a、ORF4b、ORF5均有一定的核苷酸替换,而ORF8b相对保守。总之,中国首例输入性MERSCoV与已发表序列相比总体表现为保守,但在部分基因序列上有一定的变异。这是中国首例输入性MERS-CoV的第一次基因分析结果报道,可为该MERS-CoV感染特点的进一步研究及相关疾病的防控提供参考。
For the first laboratory-confirmed imported MERS-CoV infection in China, nasopharyngeal swab samples were used for nucleic acid extraction, gene amplification and sequencing to obtain the MERS-CoV_ChinaGD01 structural protein and accessory protein coding genes, including S gene, E gene, M gene, N gene, ORF3, ORF4a, ORF4b, ORF5 and ORF8b gene sequences. According to the evolution analysis of S gene, MERS-CoV in Chinese imported cases is similar to the Saudi endemic strains in recent years although it has a few loci variation, belonging to the nucleotide sequence variation of MERS-CoV subtype 5, N, E, M and other structural genes less. Ancillary protein evolution analysis showed that: ORF3, ORF4a, ORF4b, ORF5 have a certain nucleotide substitution, and ORF8b relatively conservative. In conclusion, the first imported MERSCoV in China was generally conserved compared to the published sequence, but some variations were found in some of the gene sequences. This is the first report of the first gene analysis of imported MERS-CoV in China, which may provide reference for the further study on the characteristics of MERS-CoV infection and prevention and control of related diseases.
