在pH 7.4、0.05mol·L~(-1) Tris-HCl缓冲液及室温条件下,用荧光光谱法、分光光度法研究了3-羟基-2-萘甲酸与人血清白蛋白(HSA)的相互作用。研究表明,3-羟基-2-萘甲酸与人血清白蛋白以摩尔比1:1结合,其结合常数为3.28×105mol~(-1)·L.根据Foerster非辐射能量转移机理,求算了给体(HSA色氨酸残基)与受体(3-羟基-2-萘甲酸)间距离r为4.74nm。
The effects of 3-hydroxy-2-naphthoic acid and human serum albumin (HSA) were studied by fluorescence spectroscopy and spectrophotometry at pH 7.4, 0.05 mol·L -1 Tris-HCl buffer and room temperature interaction. The results showed that the binding constant of 3-hydroxy-2-naphthoic acid to human serum albumin was 1: 1 and the binding constant was 3.28 × 105mol -1. According to the Foerster non-radiative energy transfer mechanism, The distance r between the donor (HSA tryptophan residue) and the acceptor (3-hydroxy-2-naphthoic acid) was 4.74 nm.
