目的探讨焦炉逸散物对焦炉工人端粒长度和端粒酶活力变化的影响。方法以焦炉作业工人215名为接触组,非焦炉作业工人125名为对照组。采空腹外周静脉血,提取外周血中白细胞基因组DNA和端粒酶,应用实时荧光定量聚合酶链式反应(PCR)检测外周血中白细胞端粒DNA相对长度、端粒酶相对活力。结果接触组外周血中白细胞端粒DNA相对长度小于对照组,端粒酶相对活力高于对照组,差异均有统计学意义(P<0.05)。接触组端粒相对长度与端粒酶活力呈明显负相关(r=-0.506,P<0.01)。炉顶工、炉底工组、炉侧工组的端粒相对长度、端粒酶活力与炉顶、炉底、炉侧的苯并芘B(a)p浓度分别呈明显负相关(r=-0.472,P<0.01)和正相关(r=0.421,P<0.01)。结论焦炉逸散物可导致端粒缩短、端粒酶活力增高,从而引起端粒稳态失衡。
Objective To investigate the effects of coke oven fugitives on telomere length and telomerase activity in coke oven workers. Methods 215 workers in the coke oven were exposed, and 125 non-coke oven workers were used as the control group. Peripheral venous blood was collected and peripheral blood leukocyte genomic DNA and telomerase were extracted. The relative leukocyte telomere length and telomerase activity in peripheral blood were detected by real-time fluorescence quantitative polymerase chain reaction (PCR). Results The relative leukocyte telomere length in the peripheral blood of the contact group was lower than that of the control group, and the relative activity of telomerase was higher than that of the control group (P <0.05). The relative telomere length in telomerase group was negatively correlated with telomerase activity (r = -0.506, P <0.01). The relative telomere length, telomerase activity and the concentration of benzopyrene B (a) p at the top, bottom and side of the furnace were negatively correlated (r = -0.472, P <0.01) and positive correlation (r = 0.421, P <0.01). Conclusion Coke oven waste can lead to shortened telomeres and increased telomerase activity, leading to the imbalance of telomere homeostasis.
