目的探讨人脐带间充质干细胞(HUCMSCs)在小鼠体内向男性生殖细胞分化的可能性。方法HUC-MSCs纯化培养后,经显微注射方法移植到不育小鼠睾丸曲细精管,对体内移植后的细胞行免疫组化、免疫荧光及激光共聚焦,观察其存活、迁移及男性生殖细胞的特异性标记物表达等情况。结果HUCMSC体内移植后免疫组化显示其在睾丸微环境中可以存活至少120 d并发生从管腔到基底膜的迁移;免疫组化、免疫荧光、激光共聚焦证明HUCMSCs移植后在睾丸微环境中可以进一步分化,表达生殖细胞特异性标记物。结论体内外定向诱导HUC-MSCs可能可以向男性生殖细胞方向分化,HUCMSCs将可能为治疗男性不育提供一种新的细胞来源,提供一条新的治疗途径。
Objective To investigate the possibility of human umbilical cord mesenchymal stem cells (HUCMSCs) differentiating into male germ cells in mice. Methods After purified and cultured, the HUC-MSCs were transplanted to the testicular seminiferous tubules of testes of mice by microinjection. Immunohistochemistry, immunofluorescence and confocal laser scanning microscope were used to observe the survival and migration of HUC-MSCs. Germ cell specific marker expression and so on. Results Immunohistochemistry showed that HUCMSC could survive for at least 120 days in the testicular microenvironment and migrate from the lumen to the basement membrane. Immunohistochemistry, immunofluorescence and laser scanning confocal microscope showed that HUCMSCs were transplanted in the testes microenvironment Can be further differentiated to express germ cell-specific markers. Conclusion HUC-MSCs may differentiate into male germ cells in vitro and in vivo. HUCMSCs may provide a new source of cells for the treatment of male infertility and provide a new therapeutic approach.
