目的检测CoC l2化学缺氧条件下,绒毛外滋养细胞中促血管生成蛋白Cyr61和NOV的转录水平表达情况,探讨其在子痫前期发病机制的研究。方法采用实时荧光定量逆转录聚合酶链反应(qRT-PCR)技术检测绒毛外滋养细胞中Cyr61和NOV mRNA的表达。结果 (1)促血管生成蛋白Cyr61和NOV在绒毛外滋养细胞中表达。(2)与正常对照组相比,缺氧条件下Cyr61和NOV转录水平降低(P<0.05)。结论 Cyr61和NOV的表达下调可能与胎盘血管生成障碍有关,从而参与了子痫前期的发病有关。
Objective To detect the transcriptional levels of Cyr61 and NOV in villous trophoblast under CoCl 2 chemical hypoxia and to explore their pathogenesis in preeclampsia. Methods Real-time fluorescent quantitative reverse transcription-polymerase chain reaction (qRT-PCR) was used to detect Cyr61 and NOV mRNA expression in trophoblast cells. Results (1) The pro-angiogenic proteins Cyr61 and NOV were expressed in extravillous trophoblasts. (2) Compared with the normal control group, the transcriptional level of Cyr61 and NOV in hypoxia decreased (P <0.05). Conclusion The down-regulation of Cyr61 and NOV may be related to placental angiogenesis disorders, which may be involved in the pathogenesis of preeclampsia.
