目的研究塞来昔布与吉西他滨(GEM)联用对Lewis肺癌C57BL/6小鼠皮下移植瘤的联合效应。方法 Lewis肺癌细胞系建立小鼠皮下移植瘤动物模型。将40只小鼠随机分为对照组(A组)、GEM组(B组)、塞来昔布组(C组)和塞来昔布与GEM联用组(D组),每组10只。C、D组自接种后第2天每日给予含塞来昔布1mg/g的饲料喂养;A、B组给予普通饲料喂养。B、D组于肿瘤长径达0.2cm时(约接种后第7天),给予GEM100mg/kg溶液每只0.2ml腹腔注射;A、C组给予等量生理盐水,每周1次,连续3周,4周后处死。实验期间观察小鼠的生存质量及肿瘤生长情况,测量肿瘤长短径并绘制肿瘤生长曲线,称量瘤重,计算抑瘤率。结果 B、C和D组小鼠移植瘤生长较A组相对缓慢。B、C和D组抑瘤率分别为54.18%、49.52%和88.59%。D组肿瘤重量明显小于A、B和C组;B组小鼠去肿瘤后体重较A、C和D组轻度减轻。结论塞来昔布和GEM均对Lewis肺癌皮下移植瘤生长具有较好的抑制作用,两者联用具有协同抗肿瘤作用,同时能够改善小鼠的生存质量。
Objective To study the combined effect of celecoxib and gemcitabine (GEM) on subcutaneous xenografts in Lewis lung carcinoma C57BL / 6 mice. Methods Lewis lung carcinoma cell lines were established subcutaneously transplanted tumor animal model. Forty mice were randomly divided into control group (group A), GEM group (group B), celecoxib group (C group) and celecoxib group with GEM (group D) . Groups C and D were fed daily with celecoxib 1 mg / g on the second day after inoculation. Groups A and B were given normal feed. Groups B and D were injected intraperitoneally with 0.2 ml of GEM100mg / kg solution when the tumor’s diameter was 0.2cm (about 7 days after inoculation); Group A and C were given normal saline once a week for 3 consecutive days Week, sacrificed after 4 weeks. During the experiment, we observed the quality of life and the growth of the mice, measured the length and breadth of the tumor, plotted the growth curve, weighed the tumor weight and calculated the tumor inhibition rate. Results The growth of transplanted tumors in mice in groups B, C and D was relatively slow compared with that in group A. The inhibition rates of B, C and D were 54.18%, 49.52% and 88.59% respectively. The weight of tumor in group D was obviously smaller than that in groups A, B and C. The weight of mice in group B was lighter than those in groups A, C and D after tumor removal. Conclusion Both celecoxib and GEM can inhibit the growth of Lewis lung carcinoma subcutaneously. The combination of both has a synergistic anti-tumor effect and can improve the quality of life of mice.
