癌的发生是一个多步骤过程,需多种基因异常的积累,其中最常异常的基因是肿瘤抑制基因P~(53)。该基因位于染色体17P,在人类许多肿瘤中其种系发育高度保守区内常发生突变。野生型p~(53)基因产物为53KD(5258.13u)的核磷酸蛋白,具有抑制肿瘤恶性表型的作用。p~(53)基因错义突变(mis-sense mutation)所产生的p~(53)蛋白在转化细胞内升高,促进细胞转化特性的表达。
The occurrence of cancer is a multi-step process that requires the accumulation of multiple gene abnormalities. The most frequently abnormal gene is the tumor suppressor gene P53. This gene is located on chromosome 17P and frequently mutates in many highly conserved germline systems in many human tumors. The wild-type p53 gene product is a 53KD (5258.13u) nuclear protein with the function of inhibiting the malignant phenotype of the tumor. The p ~ (53) protein produced by p ~ (53) mis-sense mutation was increased in transformed cells and promoted the expression of cell transformation features.
