目的:进行不同产地连翘挥发油的主要成分气相测定及其体外抗菌作用的比较研究。方法:采用水蒸气蒸馏法提取不同产地16批连翘果实中的挥发油,利用气相色谱分析检测其主要成分及含量,采用K-B纸片扩散法比较分析其抗菌活性。色谱条件为HP-5毛细管色谱柱(0.25 mm×30 m,0.25μm),程序升温为初始温度50℃,以5℃·min~(- 1)的速率升至86℃,保持3 min,再以20℃·min~(- 1)的速率升至220℃保持1 min,进样口温度230℃,检测器温度250℃。结果:α-蒎烯,莰烯,β-蒎烯,月桂烯,对伞花烃,柠檬烯和α-松油醇的线性关系良好(r均>0.999 7),线性范围分别为0.016 5~0.988 8,0.005~0.300,0.038 1~2.286 0,0.003 2~0.192 0,0.000 8~0.048 0,0.008 2~0.492 0,0.002 4~0.144 0μg;稳定性、重复性试验的RSD均<3.0%;加样回收率为99.0%~103.4%(RSD<3.0%);不同产地连翘挥发油中主要成分含量有差异,其对5种试验菌均有不同程度抑制作用。结论:所建立的连翘挥发油多成分GC测定方法简便、准确,可用于连翘药材的质量评价;抗菌实验结果可靠,可用于连翘药材的抗菌作用评价。
OBJECTIVE: To determine the main components of volatile oil from Forsythia suspense in different habitats and compare their antimicrobial activities in vitro. Methods: Volatile oil was extracted from 16 batches of Forsythia suspensa from different habitats by steam distillation. The main constituents and contents were determined by gas chromatography. The antibacterial activity was analyzed by K-B disk diffusion method. The chromatographic conditions were HP-5 capillary column (0.25 mm × 30 μm, 0.25 μm). The programmed temperature was raised to 50 ℃ and raised to 86 ℃ at 5 ℃ · min ~ (-1) for 3 min The temperature was raised to 220 ° C for 1 min at a rate of 20 ° C · min -1 and the inlet temperature was 230 ° C and the detector temperature was 250 ° C. The main constituents in the volatile oil of Forsythia suspensa from different habitats were different, which showed different degrees of inhibition on the five tested bacteria. Conclusion: The established GC method for determination of volatile oil in Forsythia suspensa is simple and accurate and can be used to evaluate the quality of Forsythia suspensa. The antibacterial test results are reliable and can be used to evaluate the antibacterial activity of Forsythia suspensa.
