为研究顺铂及同时合并43℃30min热疗作用下胃癌细胞的凋亡过程,采用细胞培养的方法,通过AO染色、DNA微电泳及电镜观察,发现人胃癌MKN 45细胞在25mg/L浓度的顺铂作用2h、43℃30min热疗作用及两者同时作用下,呈现不同程度的凋亡。电镜观察到凋亡的细胞表;现DNA微电泳呈拖尾现象,提示凋亡细胞内DNA片断化;凋亡细胞AO染色呈阳性反应,且顺铂与热疗共同作用时AO阳性率高于两者单独作用时,有显著性差异(P<0.05),但两者单独作用时AO染色比较无显著性差异。提示热疗与顺铂合用时对人胃癌细胞的凋亡具有协同作用。
In order to study the apoptosis of gastric cancer cells treated with cisplatin and at the same time at 43°C for 30 min, the cell culture method was used to observe the expression of human gastric cancer MKN 45 cells at a concentration of 25 mg/L by AO staining, DNA microelectrophoresis and electron microscopy. The effects of cisplatin for 2h, 43°C for 30min and hyperthermia both showed different degrees of apoptosis. Electron microscopic observation of the apoptotic cell surface; DNA microelectrophoresis showed a tailing phenomenon, suggesting apoptotic DNA fragmentation; apoptotic cells AO staining was a positive reaction, and AO positive rate was higher when cisplatin and hyperthermia together There was a significant difference between the two when they were used alone (P<0.05), but there was no significant difference in AO staining between the two. It is suggested that hyperthermia combined with cisplatin has a synergistic effect on the apoptosis of human gastric cancer cells.
