目的 :探讨不同细胞因子对大肠癌细胞 Fas配体 (Fas L )表达水平的调控作用。方法 :采用 Western印迹法测定了两种细胞因子 (1× 10 6 U / L TNF-α或 IFN-γ)处理后大肠癌细胞 Fas L表达水平的变化。结果 :所检测的 6株大肠癌细胞全部表达 Fas L蛋白 ;TNF-α和 IFN-γ处理 DL D- 1细胞 10 h,显著上调其 Fas L蛋白的表达水平 ;TNF-α和 IFN-γ以及 PMA +离子霉素 (ionomycin)处理 SW6 2 0细胞 10 h,也显著上调 SW6 2 0细胞的 Fas L蛋白表达水平。 TNF-α和 PMA +离子霉素处理SW6 2 0细胞 48h进一步增加 SW6 2 0的 Fas L 蛋白表达 ,IFN- γ处理 SM6 2 0则表达增加不明显。结论 :TNF- α和 IFN- γ等细胞因子具有上调某些大肠癌细胞 Fas L 表达的作用
Objective: To investigate the regulation of Fas ligand (Fas L) expression in colorectal cancer by different cytokines. METHODS: The expression of Fas L in colorectal cancer cells treated with two cytokines (1×10 6 U/L TNF-α or IFN-γ) was determined by Western blotting. Results: All 6 colorectal cancer cells were detected to express Fas L protein; TNF-α and IFN-γ treatment of DL D-1 cells for 10 h significantly up-regulated the expression of Fas L protein; TNF-α and IFN-γ and Treatment of SW6 cells with PMA + ionomycin for 10 h also significantly up-regulated the expression of Fas L protein in SW6 2 0 cells. The SW6 2 0 cells treated with TNF-α and PMA + ionomycin for 48 h further increased the Fas L protein expression of SW6 2 0 , while the expression of SM6 200 in IFN-γ treated cells was not significantly increased. Conclusion: The cytokines such as TNF-α and IFN-γ have the effect of up-regulating the expression of Fas L in some colorectal cancer cells.
