目的:探讨积雪草酸对慢性髓系白血病细胞急变株K562增殖抑制,诱导凋亡的作用及其机制。方法:四甲基偶氮唑盐(MTT)法检测细胞增殖活性,Annexin-V/PI双标流式细胞术检测细胞的凋亡率,Hoechesst33258染色法检测细胞的形态学变化,RT-PCR检测积雪草酸对survivin和bcl-2mRNA表达的影响。结果:积雪草酸对K562细胞有明显的增殖抑制作用,尤其在30μmol.L-1以上增殖抑制效应更加显著。24,48h的IC50值分别为(36.2±4.7)μmol.L-1、(27.4±3.7)μmol.L-1。积雪草酸能诱导细胞发生凋亡,可见典型的凋亡形态,呈剂量依赖性。积雪草酸可使survivin、bcl-2mRNA的表达呈剂量依赖性降低。结论:积雪草酸能抑制K562细胞的增殖及诱导凋亡,呈剂量依赖性。积雪草酸的促凋亡机制可能与下调survivin和bcl-2的转录水平有关。
OBJECTIVE: To investigate the effect of Asiatic acid on the proliferation inhibition, apoptosis induction and its mechanism of acute myeloid leukemia cell line K562. Methods: MTT assay was used to detect cell proliferation activity. Annexin-V/PI double-label flow cytometry was used to detect the apoptosis rate of the cells. Hoechesst 33258 staining was used to detect the morphological changes of the cells. RT-PCR detection was performed. Effect of Asiatic acid on the expression of survivin and . RESULTS: Asiatic acid significantly inhibited the proliferation of K562 cells, especially at 30 μmol.L-1. The IC50 values at 24 and 48 h were (36.2±4.7) μmol.L-1 and (27.4±3.7) μmol.L-1, respectively. Asiatic acid can induce apoptosis in cells, showing typical apoptotic morphology in a dose-dependent manner. Asiatic acid decreased the expression of survivin and bcl-2 mRNA in a dose-dependent manner. Conclusion: Asiatic acid can inhibit the proliferation and induce apoptosis of K562 cells in a dose-dependent manner. The pro-apoptotic mechanism of Asiatic acid may be related to the down-regulation of the expression of survivin and bcl-2.
