为研究光照周期对黄鳝不同组织的基因组DNA甲基化的影响,设置3个不同的光照时间(8、10和12h)处理,对黄鳝心、肝、脾、肾、肠、肌肉和脑等组织基因组DNA甲基化用甲基化敏感扩增多态性(methylation sensitive amplified polymorphism,MSAP)技术进行分析。结果显示,不同组织基因组DNA对光照时间有不同的反应。其中,肌肉基因组DNA甲基化对光照周期的变化较为敏感,不同光照周期下肌肉基因组DNA总甲基化差异显著,光照周期为12h半甲基化和其他处理相比差异也很显著;肾脏和脑的基因组DNA甲基化对光照周期变化最不敏感,各处理组之间全甲基化、半甲基化和总甲基化差异都不显著;肝脏、肠、脾和心脏表现比较复杂,其中肝脏不同处理下基因组DNA半甲基化和总甲基化差异都不显著,光照周期为8h时,全甲基化差异显著;肠不同处理下基因组DNA全甲基化、半甲基化差异不显著,在光照周期为10h的处理组的总甲基化和其他处理组差异显著;脾脏在光照周期为10h和8h时全甲基化和总甲基化各处理组差异显著,在光照周期为12h时半甲基化和其他处理组差异显著;心脏在光照周期为10h时半甲基化和其他处理组差异显著。

The liver, intestine, spleen and heart were more complicated, There was no significant difference in the methylation and total methylation of genomic DNA under different treatments in the liver. There was significant difference in total methylation when the photoperiod was 8h.
