目的了解三七总甙(PNS)对间质纤维化人肾成纤维细胞凋亡的影响及其分子机理,探索治疗肾间质纤维化的有效药物。方法在成功分离培养人肾间质成纤维细胞基础上,通过dUTP标记DNA断裂点显色及流式细胞仪等方法,观察PNS对细胞凋亡及其调控蛋白CMyc、Fas表达的影响。结果PNS明显促进细胞凋亡,导致细胞数量减少;细胞CMyc蛋白表达水平升高,而Fas蛋白的表达不受PNS的影响。结论PNS通过诱导CMyc蛋白表达上调,促进人肾间质成纤维细胞凋亡,使细胞生存数量下降,可能是治疗肾间质纤维化的有效药物。
Objective To investigate the effect of Panax notoginseng saponins (PNS) on the apoptosis of human renal fibroblasts induced by interstitial fibrosis and its molecular mechanism, and to explore effective drugs for the treatment of renal interstitial fibrosis. Methods On the basis of successful isolation and culture of human renal interstitial fibroblasts, the effects of PNS on the apoptosis and the expression of regulatory proteins C-Myc and Fas were observed by dUTP-labeled DNA breakpoint coloration and flow cytometry. Results PNS significantly promoted apoptosis and led to a decrease in the number of cells; the expression of C-Myc protein was increased, and the expression of Fas protein was not affected by PNS. Conclusion PNS can increase the expression of CMyc protein, promote the apoptosis of human renal interstitial fibroblasts, and decrease the number of cells. It may be an effective drug for the treatment of renal interstitial fibrosis.
