目的本研究探讨肝癌组织中miR-370的表达,分析其与临床病理和预后的关系。方法收集90对肝癌组织和相应的癌旁组织标本,20例正常肝组织标本为对照,分析不同组织中miR-370的表达水平与临床病理指标之间的关系,并对其中40例随访病例进行了生存分析。结果 Real-time PCR结果表明与正常肝组织相比,miR-370在癌旁组织中表达显著下降,在肝癌组织的表达进一步下降。对90对标本对应病例的临床资料的分析,结果显示miR-370的表达与肿瘤大小、TMN分期、是否血管侵犯、有无肿瘤卫星灶、是否包膜侵犯等肝癌恶性指标密切相关。Kaplan-Meier生存分析表明,低miR-370表达的肝癌患者总体生存时间短于高表达者,结果有统计学意义。结论 miR-370在肝癌组织中的表达与正常肝组织相比显著下调,miR-370低表达水平与肿瘤恶性程度和预后密切相关。
Objective To investigate the expression of miR-370 in hepatocellular carcinoma (HCC) and to analyze its relationship with clinicopathology and prognosis. Methods Totally 90 specimens of HCC tissues and corresponding adjacent tissues were collected and 20 specimens of normal liver tissues were collected as controls. The relationship between the expression of miR-370 and clinicopathological parameters in different tissues was analyzed. Forty cases were followed up Survival analysis. Results Real-time PCR results showed that compared with normal liver tissue, miR-370 expression decreased significantly in paracancerous tissues and further decreased in HCC tissues. The analysis of the clinical data of 90 pairs of cases showed that the expression of miR-370 was closely related to the malignant indexes of liver cancer, such as tumor size, TMN stage, whether vascular invasion, tumor satellites, or capsule invasion. Kaplan-Meier survival analysis showed that the overall survival time of HCC patients with low miR-370 expression was shorter than that of high expression, with statistical significance. Conclusion The expression of miR-370 in hepatocellular carcinoma is significantly down-regulated compared with normal liver tissue. The low miR-370 expression is closely related to the malignancy and prognosis of HCC.
